Research Overview
Inhibiting methionine aminopeptidase 2 is a recognized strategy for suppressing angiogenesis and cancer progression. This study synthesized small-molecule fumagillol derivatives carrying adamantane side groups and evaluated them for inhibitory activity, identifying a lead compound designated AD-3281.
AD-3281 effectively suppressed proliferation of both cancer and endothelial cells and impaired endothelial tube formation in vitro. Administered systemically, it was well tolerated and significantly suppressed human melanoma and mammary tumor xenografts.
