Research Overview

3D culture models have become biomimetic alternatives for initial drug screening because they reproduce key aspects of the tumor microenvironment: 3D architecture and the interactions of tumor cells with the extracellular matrix and surrounding non-tumor cells. Accurate 3D models can also reduce animal use, in line with the European Union’s 3R principle of replacement, reduction, and refinement.

This review examines the impact of 3D culture models on cancer research — their advantages, their limitations, and how compatible they are with high-throughput screening and automated systems.

Key Discoveries

  • Reviews 3D culture models as biomimetic alternatives for early cancer drug screening
  • Frames accurate 3D models as a route to reducing laboratory animal use under the 3R rule
  • Assesses compatibility with high-throughput screening and automation
