Research Overview

This study compared how doxorubicin and 5-fluorouracil act in combination against breast cancer, testing both MCF-7 cells and cancer stem cells isolated from that line, in 2D and 3D culture.

After isolating and characterizing the cancer stem cells — confirming they express stemness factors — the antiproliferative effects of each drug were measured in 2D and combination index values calculated to determine synergy or antagonism. 3D tumor spheroids served as the second model, and qRT-PCR analysis examined apoptosis-related gene expression, letting the authors compare drug response between the two culture formats and between bulk tumor cells and their stem-like subpopulation.
