Drug combinations for pancreatic cancer are usually assessed in 2D culture. This study compared the therapeutic effect and synergistic potential of a doxorubicin-resveratrol combination in 2D monolayers versus 3D spheroids of PANC-1 pancreatic cancer cells — the first such direct comparison for this pairing.
Molar ratios from 5:1 to 1:5 were tested. Viability was most affected by combinations containing higher resveratrol content (1:2 to 1:5 ratios), a result attributed to resveratrol’s own activity — and the 2D-versus-3D comparison shows how much the culture format shapes apparent combination efficacy.
