Research Overview

Tissue mimics several hundred microns across are a valuable culture format for engineered tissue, but their density and size make them hard to observe optically. Light sheet fluorescence microscopy (LSFM), an emerging technique for 3D optical sectioning of large samples, appears well suited to them.

This study compared the effectiveness of different light sheet illumination modalities reported in the literature for improving resolution and/or reducing light exposure in complex 3D samples. To make the comparison rigorous and fair, the authors also developed a systematic, computerized benchmarking method for evaluating the modalities.
